Scientific Basis: How Enterolert® 250 Works

 

Enterolert 250 uses a Defined Substrate Technology® (DST®) nutrient indicator to detect enterococci. Enterolert 250 provides confirmed presence/absence results in 24 hours. Enterolert 250 utilises orth-nitrophenyl-ß-D-glucoside as a nutrient indicator and incorporates a specifically designed blue background colour in its formulation. When the substrate is metabolised by enterococci, the sample turns from blue to green to indicate detection. Any change from the original colour to green is considered a positive result. No ultraviolet light source is required. Enterolert 250 detects enterococci at 1 cfu in 250 mL bottled water samples in 24 hours.

How does the product work?

Enterolert 250 is designed to detect Enterococcus species in bottled water samples. It is based on IDEXX’s patented Defined Substrate Technology® (DST®). Enterolert 250 utilizes ortho-nitrophenyl-ß-D-glucoside as a nutrient indicator and incorporates a specifically designed blue background colour in its formulation. When the enzyme substrate is metabolized by the enterococci, the sample turns from blue to green to indicate a positive detection. Any change from the original colour to green is considered a positive result. No ultraviolet light source is required. Enterolert 250 detects enterococci at 1 cfu in 250 mL bottled water samples in 24 hours.

 

What species of enterococci can Enterolert 250 detect?

Enterolert 250 is designed to detect enterococci in bottled water. As with any microbial method, some Enterococcus species are more easily detected than others. Some examples of Enterococcus species we have detected in trials and studies are:

Enterococcus faecium
Enterococcus hirae
Enterococcus gallinarum
Enterococcus casseliflavus
Enterococcus durans
Enterococcus avium

 

Is there a comparator for Enterolert 250?

There is no comparator for Enterolert 250. For comparison, a negative control can be used when interpreting results.

 

What is the incubation time and temperature of Enterolert 250?

Enterolert 250 is incubated at 41°C ± 1.0°C for 24 hours and the results are valid up to 28 hours, unless your local regulations indicate differently.

 

Are results valid prior to 24 hours?

Yes, if they are positive. An inoculated Enterolert 250 sample that is positive for enterococci before 24 hours is a confirmed positive test for enterococci.

 

Are results valid after 28 hours?

Yes, if they are negative. If an inoculated Enterolert 250 sample is inadvertently incubated over 28 hours, lack of green colour is a valid negative test; a green colour after the 28 hours is not valid and should be repeated or verified.

 

What are the recommended storage conditions for Enterolert 250?

Store Enterolert 250 at 2°C–25°C, away from direct light and humidity.

 

What is the normal colour change that should take place with Enterolert 250?

When the enzyme substrate is metabolised by the enterococci, the sample turns from blue to green to indicate a positive result. Any change from the original colour to green is considered a positive result.

 

What water types can the product be used on/what water type is the product designed to be used for?

Enterolert 250 is designed specifically for bottled water testing.

 

What colour and consistency should the Enterolert 250 reagent powder be?
